Clearing a lot you intend to build on
On a building lot the customer for the finished ground is not you — it is a builder with a drawing and an inspector. That changes what "cleared" has to mean and how much of it can be left to judgement.

The short answer
A build requires below-grade removal within the footprint and its working margin, because root plates decay and foundations do not tolerate the voids that follow.
It also requires that debris does not remain in the ground anywhere a structure, a drive, or a service trench will sit. Burying material on site is the single most damaging shortcut available on a building lot, and it is not visible once the machine has left.
Topsoil is worth separating and stockpiling rather than mixing. It is the most valuable material on the site for later landscaping, and it is the easiest to lose permanently.
Clearing does not produce a buildable pad. Forming ground to a tolerance is a separate trade, and it happens after backfill has settled.

What has to be cleared, and how far beyond the footprint
Clearing exactly the footprint is the most common scoping error on a build, because nobody builds a structure from inside its own outline.

Why buried debris is the failure that surfaces years later
This is worth being blunt about, because it is invisible at handover and expensive afterwards.
Woody material buried in fill decays. As it does, the volume it occupied becomes void, and whatever sits above settles into it. Under a lawn that is a dip. Under a slab, a drive, or a footing, it is a structural defect that is discovered by cracking.
The economic pressure to bury is real: it removes the disposal cost and the haulage entirely. That is precisely why the scope document should state where material goes, and why a discounted price on a lot with no visible debris pile deserves a direct question about the destination.
Topsoil, and the money most owners throw away
The upper soil layer on a wooded lot is the accumulation of decades of leaf fall, and it is the material a landscaper would otherwise sell you back.
Stripping and stockpiling it before earthworks begin costs a little machine time and a corner of the site. Mixing it into subsoil during grubbing costs nothing at the time and everything later, because there is no way to separate it again.
A stockpile needs to be somewhere it will not be driven over, and it should not be left bare through a wet season on a slope. Both of those are cheap to arrange if decided in advance and impossible to arrange afterwards.
Where clearing stops and site preparation begins
The handover point between these two trades is where build projects most often lose time.
Clearing ends with the standing growth gone, the material dealt with, the stumps out where required, and the holes backfilled. The ground at that point is uneven, loose in places, and still settling. It is not a pad.
Site preparation takes that ground and forms it: cut and fill to a design, compaction in layers, a surface within a tolerance somebody will measure. It cannot sensibly begin until backfill has settled, and it cannot be done well in saturated conditions.
Where the same contractor does both, ask how long they leave between them. Where two contractors are involved, that gap is your scheduling problem and worth agreeing early.
Keeping trees deliberately rather than by accident
Mature trees on a lot are worth more than they cost to protect, and they are routinely lost for want of an hour of planning.
Retention needs three things: a decision about which trees, physical marking on the ground, and a protected zone the machines do not enter. Root damage from repeated traffic within the canopy spread is the usual cause of loss, and it kills trees slowly enough that nobody connects the cause to the effect.
Be realistic about which trees can survive. A tree left standing alone after the shelter around it has gone is exposed to wind it never had to resist, and species vary in how well they tolerate that. Ask the contractor which of your candidates they would actually expect to keep.

Questions people actually ask
Does clearing include the driveway route?
Only if you say so. It is frequently excluded because it is a corridor rather than an area and is often priced separately. Since the route has to carry loaded vehicles through the build, it is worth including deliberately rather than discovering later.
Can material be buried in an area away from the house?
Sometimes, where local rules allow it and where nothing will ever be founded, driven, or dug there. It still creates a settling area and it still has to be recorded, otherwise a future owner discovers it. Ask what your jurisdiction permits before assuming.
Should the soil test happen before or after clearing?
That depends what the test is for and who requires it — some assessments need undisturbed ground and some need access. Ask whoever requires it, and ask before anything is disturbed, because undisturbed ground is the one condition you cannot recreate.
How much margin does a builder need around the footprint?
It varies with the structure and the method, so the builder is the right person to ask, not the clearing contractor. Getting that answer before quoting avoids the most common second-mobilisation charge on build sites.
